واکاوی بیوانفورماتیکی و فیلوژنی ناحیه 5`UTR ژن نوروپپتید وای (Neuropeptide Y) و ارتباط آن با صفات تولیدی در مرغ‌ بومی فارس

چکیده
این پژوهش به­منظور بررسی فیلوژنی و بیوانفورماتیکی جهش تک‌نوکلئوتیدی در ناحیه 5`UTR ژن نوروپپتید وای (Neuropeptide Y=NPY) و ارتباط آن با صفات تولید تخم­مرغ و وزن بدن در مرغان بومی استان فارس، انجام گرفت. از 200  قطعه مرغ بومی ایستگاه پرورش و اصلاح­نژاد مرغ بومی استان فارس خون­گیری انجام شد. استخراج DNA با روش نمکی انجام شد. جهش تک‌نوکلئوتیدی ژن NPY با تکنیک PCR-RFLP شناسایی شد. فراوانی آلل‌های A و B به­ترتیب 46/0 و 54/0 برآورد شد و جمعیت مورد مطالعه در تعادل هاردی-واینبرگ نبود. واکاوی آماری نشان داد که، بین چندریختی نوروپپتید وای و صفات مورد مطالعه (شمار تخم­مرغ، اولین سن تخم­گذاری­، وزن تخم­مرغ در 84 هفتگی و وزن بدن در 12 هفتگی) ارتباط معنی­داری وجود ندارد. همچنین، واکاوی فیلوژنی نشان داد که ناحیه توالی‌یابی­شده ژن NPY مرغ بومی فارس با اطلاعات توالی‌های EU447658.1 و EU447659.1 دارای بیشترین شباهت است. نکته قابل توجه در واکاوی فیلوژنی، وجود ژن NPY از گونه بلدرچین بود که می‌تواند نشان دهنده نقش کلیدی این ژن در تنظیم فرآیندهای فیزیولوژیک محسوب گردد.

چکیده English

The aim of this study was bioinformatics and phylogenetic analysis for 5′UTR region of neuropeptideY (NPY) gene and its association with body weight and egg production traits in Fars native chickens. For this porpuse 200 blood samples were collected from Fars Native chickens Breeding Center (Shamsabad station). The whole genome DNA was extracted using salting out protocol. The promotor region was amplified by PCR and SNP genotyping was performed by RFLP method. The frequencies of alleles A and B were estimated as 0.46 and 0.54, respectively. The population was deviated from H-W equilibrium. Statistical analysis indicated that there is no significant association between NPY genotypes and studied traits (egg number, age at first laying, egg weight at 84 weeks and body weight at 12 weeks). The results of phylogenetic analysis showed that the sequences of the NPY gene of Fars native chickens has the most simalirity with EU447658.1andEU447659.1 sequence information. Interstingly, there is NPY gene of quail in phylogenetic analysis which can show the key role of NPY gene for regulating the physiological process.
