جستجوی ساختار علّی بین صفات گوسالهزایی در نخستین زایش گاوهای هلشتاین ایران
در این پژوهش با استفاده از دادههای گردآوریشدۀ مربوط به صفات گوسالهزایی شامل سختزایی، وزن تولّد گوساله و طول دورۀ آبستنی در نخستین زایش 29950 رأس گاو هلشتاین ایران که در سالهای 1383 تا 1393 در 131 گلۀ تحت پوشش مرکز اصلاح نژاد و بهبود تولیدات دامی کشور گردآوریشده بودند، ساختار علّی بین صفات گوسالهزایی با استفاده از الگوریتم جستجوی IC بررسی شد. در ساختار علّی شناساییشده ضریبهای ساختاری مربوط به تأثیر علّی صفات وزن تولّد گوساله (002/0±060/0) و طول دورۀ آبستنی (002/0±007/0) بر صفت سختزایی معنیدار بودند. تأثیر طول دورۀ آبستنی بر وزن تولّد گوساله (005/0±219/0) معنیدار بود. با در نظر گرفتن این ساختار علّی یک مدل سه صفتی یکسویه مبتنی بر مدلهای معادلههای ساختاری تدوین و با استفاده از معیار انحراف اطلاعات (DIC) و قابلیت پیشبینی (ارزیابیشده با معیارهای میانگین مربعات خطا و همبستگی بین مقادیر پیشبینیشده و مشاهدهشده) با مدل سه صفتی استاندارد متناظر مقایسه شد. نتایج نشان دادند، صفت سختزایی تحت تأثیر علّی وزن تولّد گوساله و طول دورۀ آبستنی گاو قرار دارد و مدل سه صفتی یکطرفه نسبت به مدل سه صفتی استاندارد برتری دارد و در نظر گرفتن رابطههای علّی یادشده در ارزیابی ژنتیکی صفات گوسالهزایی اهمیت دارد.

ارزیابی انفرادی و ترکیبی قابلیت هضم و تخمیر گیاهان شورزی سلمکی ساقهسفید، سیاهشور و اشنان در شترهای یک کوهانه
در این مطالعه تخمیرپذیری سه گونه گیاهی شورزی تحت چرای شترهای تک کوهانه بنامهای سلمکی ساقه سفید، اشنان و سیاه شور بهصورت انفرادی و با جایگزینی 0، 5/33، 5/66 و 100 درصد با یکدیگر در قالب طرح کامل تصادفی با نه تیمار آزمایشی توسط روش تولید گاز ارزیابی شد. نمونههای گیاهی در طول فصل چرای پاییزه و زمستانه از مراتع جنوب استان خوزستان گردآوری شدند. جیرههای مورد بررسی تفاوت معنیداری از نظر سرعت و میزان گاز تولید شده داشتند (05/0>P). سلمکی ساقه سفید (T2) بهطور معنیداری کمترین میزان تولید گاز را داشت و در مقابل جیرۀ حاوی 5/66 درصد سیاه شور + 5/33 درصد اشنان (T8) بیشترین میزان تولید گاز را داشت (05/0>P). سلمکی ساقه سفید بهطور معنیداری کمترین و اشنان بیشترین قابلیت هضم حقیقی، مادۀ آلی قابل تخمیر، کل اسیدهای چرب فرار تولیدی، انرژی قابل سوختوساز (متابولیسم) و تولید تودۀ میکروبی را داشتند. کمترین انرژی قابل سوختوساز در تیمار T2مشاهده شد. بازدۀ تولید توده میکروبی در گیاه سلمکی ساقه سفید بالاتر از دیگر جیرهها بود و پایینترین بازده متعلق به جیرۀ T8 بود. میزان نیتروژن آمونیاکی تولید شده در همۀ جیرههای مورد بررسی بالاتر از سطح بحرانی آنها بوده و میتوانند مقادیر کافی از نیتروژن را برای بیشترین تولید تودۀ میکروبی تأمین کنند. بنابراین، گیاه اشنان و جیرۀ T8 بهترین جیره برای تأمین نیازهای مواد مغذی شتر هستند؛ اما با دیدگاه احیاء مرتع، جیرههای ترکیبی برتر از هرکدام از سه گیاه بهتنهایی هستند.

تأثیر افزودن سرما محافظهای نفوذی گلیسرول، دی متیل استامید و دی متیل فرمامید و غیر نفوذی ترههالوز و ساکارز بر انجمادپذیری اسپرم اسب
این پژوهش بهمنظور مقایسۀ تأثیر افزودن سطوح مختلف سه نوع سرمامحافظ نفوذی (گلیسرول، دی متیل استامید، دی متیل فرمامید) و دو نوع سرمامحافظ غیرنفوذی (ترههالوزو ساکارز) به رقیقکنندۀ اسپرم اسبهای ترکمن انجام شد. برای این منظور چهار سر نریان بالغ ترکمن برای اسپرمگیری به کار گرفته شد. پس از گرفتن مایع منی و فرآوری آن بهمنظور جداسازی ژل و پلاسمای منی در آزمایشگاه، نمونههای منی به گروههای تیماری شامل سطوح 5 و 7 درصد سرمامحافظهای نفوذی گلیسرول، دی متیل استامید (DMA) و دی متیل فرمامید (DMF) و سطوح 50 و 100 میلی مول سرمامحافظهای غیرنفوذی ترههالوز و ساکارز حلشده در رقیقکننده تقسیم و با یک روش استاندارد، منجمد شدند. پس از یخگشایی، فراسنجههای جنبایی و زندهمانی برای همۀ تیمارها ارزیابی شد. نتایج این پژوهش نشان داد، زندهمانی اسپرمهای منجمدشده با رقیقکنندۀ حاوی 5 یا 7 درصد DMF (بهترتیب 93/0±6/68 و 22/2±2/63 درصد) و سطوح 5 یا 7 درصد DMA (به ترتیب 69/1±6/68 و 76/2±6/63 درصد) از اسپرمهای منجمدشده با رقیقکنندۀ حاوی سطوح 5 یا 7 درصد گلیسرول (به ترتیب 06/3±8/56 و 03/1±6/55 درصد) بهطور معنیداری بهتر بود (05/0P<). جنبایی کل اسپرمهای تیمارشده با سطح 5 درصد DMA (02/1±4/68 درصد) نسبت به سطح 7 درصد DMF (46/1±8/61 درصد) بهطور معنیداری افزایش نشان داد (05/0P<). همچنین زندهمانی و جنبایی اسپرمها تحت تأثیر نوع سرما محافظهای غیر نفوذی قرار نگرفته و این سرما محافظها تفاوت معنیداری نسبت به هم نشان ندادند (05/0P>). بر پایۀ این نتایج میتوان گفت که سرمامحافظهایی مانند DMA و DMF میتوانند جایگزین مناسبی برای گلیسرول باشند.

تأثیر پنج روش مختلف همزمانی فحلی بر قطر فولیکول تخمکریزیکننده، غلظت پروژسترون پس از تلقیح و درصد گیرایی گاو هلشتاین در تنش حرارتی
هدف از این پژوهش، مطالعه تأثیر پنج روش همزمانی فحلی بر قطر فولیکول تخمکریزیکننده، غلظت پروژسترون در زمان تلقیح و روزهای 5 و 12 پس از تلقیح و درصد گیرایی در شرایط تنش حرارتی در گاو هلشتاین بود. بدین منظور 250 رأس گاو هلشتاین (میانگین تولید روزانه بیشتر از 30 کیلوگرم در روز و نوبت زایش 1/1±6/3) بهطور تصادفی انتخاب و در گروههای آزمایشی زیر شامل 1- آوسینک (OV)، 2- پری سینک/ آوسینک (PO)، 3- دابل آوسینک (DO)، 4- G7G آوسینک (G7G)، 5- PG7G آوسینک(PG7G) قرار گرفتند. نتایج نشان داد، قطر فولیکول تخمکریزیکننده در گاوهایی که با روش OV همزمان شدند در مقایسه با دیگر روشها بهطور معنیداری کوچکتر بود. غلظت پروژسترون در روزهای 5 و 12 پس از تلقیح در گاوهایی که با روش OV همزمان شده بودند در مقایسه با دیگر روشها کمتر بود (05/0 >P). گاوهایی که با روش OV همزمان شده بودند در مقایسه با دیگر روشها کمترین درصد گیرایی را در روز 32 و 60 پس از تلقیح داشتند (05/0 >P). بین دیگر روشهای همزمانی تفاوتی در درصد گیرایی در روزهای 32 و 60 پس از تلقیح وجود نداشت. بهطورکلی نتایج این پژوهش نشان داد، استفاده از روشهای بر پایۀ پیشهمزمانی پیش از آغاز آوسینک میتواند سبب افزایش باروری در گاوهای هلشتاین در شرایط تنش حرارتی شود.

ارزیابی بیان ژنهای مؤثر بر رشد و نمو غدۀ پستان گاو با مصرف روغن ماهی
این مطالعه بهمنظور ارزیابی تأثیر مصرف روغن ماهی در مقایسه با روغن پالم بر بیان ژنهای مؤثر بر رشد بافت پستان گاوهای هلشتاین انجام شد. ده رأس گاو با دستکم یک شکم زایش بهطور تصادفی از 42 روز پیش از تاریخ احتمالی زایش تا 63 روز پس از آن به دو گروه روغن ماهی (FO) و گروه روغن پالم (PO) اختصاص یافتند. خونگیری با فاصلۀ 21 روز و نمونهگیری از بافت پستان با فاصلۀ دو ماهه (روزهای 7 و 63 پس از زایش) انجام شد. تفاوت معنیداری بین دو گروه از نظر غلظت استرادیول و پروژسترون سرم خون وجود نداشت. نتایج بررسی بیان ژن نشان داد، بیان نسبی ژن IGF-Iدر گروه روغن ماهی کاهش (05/0P<) و بیان نسبی ژن TNF-α تمایل به کاهش داشت (06/0P=). بیان ژن Bcl-2 در گروه روغن ماهی در طول زمان تمایل به افزایش داشت (10/0P=). نسبت بیان ژن BCL-2/ BAX با نزدیک شدن به اوج تولید شیر، برای گروه روغن پالم روندی کاهشی نشان داد، اما برای گروه روغن ماهی روندی افزایشی داشت که نشانۀ کاهش خودکشی یاختهای یا پسرفت (آپوپتوزیس) در بافت پستان است. بنابراین، احتمال میرود که مصرف روغن ماهی در جیرۀ گاوهای شیری هلشتاین از نیمههای دورۀ خشکی تا اوج شیردهی، سبب به تعویق انداختن روند آپوپتوزیس پستان شود.

در این پژوهش توارث سیتوپلاسمی وزن بیدۀ یکسالگی در بز نژاد مرخز با استفاده از روش آماری بیزی بررسی شد. برای برآورد فراسنجه (پارامتر)های ژنتیکی از رکوردهای گردآوریشده در سالهای 1390-1371 در ایستگاه پژوهشی اصلاح نژاد بز مرخز در سنندج استفاده شد. برای بررسی اثر عاملهای محیطی بر دادههای این صفت از رویۀ GLM نرمافزار آماری SAS و برای برآورد فراسنجههای ژنتیکی از نرمافزار Gibbs1f90، بر پایۀ مدل دام از رویۀ نمونهگیری گیبس، استفاده شد. عاملهایی مانند سال تولد، جنس و سن مادر بهعنوان اثر ثابت و سن دام و وزن بدن در هنگام رکورد برداری بهعنوان متغیرهای کمکی در مدل گنجانده شدند. بنابر نتایج بهدستآمده از این پژوهش، بهترین مدل انتخابشده برای صفت وزن بیده، مدل دربرگیرندۀ اثر ژنتیکی افزایشی مستقیم، ژنتیکی افزایشی مادری، محیط دائمی مادری و اثر ژنتیکی سیتوپلاسمی با در نظر گرفتن کوواریانس بین اثر ژنتیکی افزایشی مستقیم و مادری بود. درصد واریانس ژنتیکی افزایشی مستقیم، ژنتیکی افزایشی مادری، محیط دائمی مادری و ژنتیکی سیتوپلاسمی به ترتیب 27/19، 6/6، 03/3 و 82/1 از کل واریانس برآورد شد. نقش اثر سیتوپلاسمی با توجه به معنیدار بودن برای ورود به مدل از یکسو و کم بودن میزان واریانس آن از سوی دیگر، بهعنوان تصحیحکننده اهمیت دارد، ولی نمیتواند بهعنوان معیار انتخاب لحاظ شود.

تولید و بهکارگیری مؤثر آنتیبادی اختصاصی زردۀ تخممرغ (IgY) برای مهار باکتری استافیلوکوکوساورئوس
این بررسی بهمنظور ارزیابی ویژگیهای ایمنی (ایمونو)شیمیایی و نیز تأثیر مهارکنندگی پودر پادتن (آنتیبادی) اختصاصی و غیراختصاصی زردۀ تخممرغ بر رشد باکتری استافیلوکوکوساورئوس، بهعنوان یکی از مهمترین سروتیپهای استافیلوکوکوس بیماریزای انسانی و دامی در شرایط برونتنی انجام شد. این آزمایش با بهرهگیری از شانزده قطعه مرغ تخمگذار لگهورن سفید سویۀ های-لاین 36W- که در سن 40 هفتگی قرار داشتند صورت گرفت. برای ایمن کردن پرندگان، پس از بررسی منحنی رشد میکروب و کشت آنها تا پایان مرحلۀ رشد تصاعدی، واکسن کشتهشده با بهرهگیری از امواج فراصوت تهیه و تزریق شد. پس از گردآوری تخممرغها در یک دورۀ زمانی 8 هفته، IgY با بهرهگیری از روش پولسون جداسازی و یخ خشکانی1 شد و در ادامه ویژگیهای ایمنیشیمیایی و ضد باکتریایی آن سنجش شد. نتایج بیانگر آن است که پس از نخستین ایمنسازی، فعالیت اختصاصی پادتن علیه باکتری مورد آزمون در پلاسمای مرغهای تخمگذار و تخممرغ افزایش یافت و این افزایش در دورۀ آزمایش در سطوح بالا حفظ شد (01/0 < P). غلظت IgY و نیز درصد خلوص آن در پودر پادتن اختصاصی 71 درصد بالاتر (05/0 < P) از پودر آنتیبادی غیراختصاصی بود. بهرهگیری از سطوح 100 و 200 میلیگرم پودر پادتن اختصاصی سبب کاهش معنیدار جمعیت باکتریایی در ساعتهای 4 و 6 نگهداری (انکوباسیون) شد. در این میان بالاترین اثر مهاری در انتهای مرحلۀ (فاز) لگاریتمی متعلق به سطح 200 میلیگرم پودر پادتن اختصاصی بود که توانست سبب کاهشlogCFU/mL 2/1 در مقایسه با گروه شاهد (کنترل) شود (01/0 < P). لذا این یافتهها بیانگر آن است که باکتری استافیلوکوکوس اورئوس بههمراه ادجوانت فروند (نوع کامل در تزریق اول و نوع ناقص در تزریق دوم)، خاصیت القایی بالایی داشته و بهرهگیری از روش پولسون راهی مؤثر برای تولید درازمدت و با بازدۀ بالا، برای به دست آوردن IgY اختصاصی با درصد خلوص و توان ضدباکتریایی (آنتیباکتریال) مطلوب است.

تأثیر اسناد کردن و تراکم نشانگری در بهبود درستی ارزیابیهای ژنگانی
با دسترسی به اطلاعات نشانگرهای تک نوکلئوتیدی (SNP)، پیشبینی شایستگی ژنتیکی افراد بدون اطلاعات پدیدگانی (فنوتیپی) با استفاده از ارزیابی ژنگانی (ژنومی) امکانپذیر شده است. لیکن، استفاده از تراشه (چیپ)های متراکم برای ارزیابی همۀ افراد جمعیت مقرونبهصرفه نیست. برای دستیابی به بیشترین بازدهی در بررسیهای ژنگانی میتوان گروهی از حیوانها را با تراشههای با تراکم بالا و دیگر افراد را با تراشههای کمتراکم تعیین نژادگان (ژنوتیپ) کرد، سپس آنها را به تراکم بالا اسناد (ایمپیوت) کرد. در این بررسی، تأثیر سه تراشۀ کمتراکم (1k، 2k و 4k)، اسنادشده به تراکم بالای 10k و رابطۀ خویشاوندی بین جمعیت مرجع و جمعیتهای تأیید بر درستی ارزیابی ژنگانی و نیز همبستگی ارزشهای اصلاحی برآوردشده در تراشههای مختلف با استفاده از دادۀ همانندسازیشده، بررسی شد. برای ساختن تراشههای کمتراکم، 10، 20 و 40 درصد از نشانگرهای 10k بهصورت تصادفی انتخاب شدند. اسناد کردن (ایمپیوتیشن) با استفاده از نرمافزار FImpute صورت گرفت. بهطورکلی با افزایش تراکم نشانگری، همبستگی بین ارزشهای اصلاحی برآوردشده بهدستآمده از تراشههای مختلف افزایش پیدا کرد. بهگونهای که، درستی ارزیابی ژنگانی تراشۀ کمتراکم 4k و تراشۀ متراکم 10k همسان بودند. همچنین پس از اسناد کردن نژادگانهای تراشۀ 4k به 10k، تفاوتی بین درستی ارزیابیهای ژنگانی ایجاد نشد. با افزایش فاصلۀ جمعیت مرجع و جمعیتهای تأیید، درستی اسناد کردن کاهش یافت.

تأثیر بنتونیت نانوساختار پیلارد بر عملکرد و فراسنجههای بیوشیمیایی خون جوجههای گوشتی چالشیافته با آفلاتوکسین
هدف از این آزمایش، بررسی تأثیر بنتونیت سدیم پیلارد (PB) بر عملکرد، وزن نسبی اجزاء لاشه و فراسنجههای بیوشیمیایی خون جوجههای گوشتی چالشیافته با آفلاتوکسینبود. بدین منظور از 128 قطعه جوجهخروس یکروزه راس 308 در یک طرح کامل تصادفی با چهار تیمار و چهار تکرار و هشت پرنده در هر تکرار به مدت 35 روز استفاده شد. تیمارهای آزمایشی شامل: شاهد منفی (C)، شاهد مثبت (جیرۀ پایه + آفلاتوکسینAFB1) و دو سطح 25/0 و 75/0 درصد بنتونیت نانو ساختار پیلارد (PB) که به جیرۀ پایۀ آلوده به آفلاتوکسینAFB1اضافه شد، میزان آفلاتوکسین در جیرههای آلوده در دورۀ آغازین و رشد برابر ppb500 و در پایانی ppb1000 بود. ستون حایل بودن (پیلارینگ) در بنتونیت سدیم باعث افزایش میزان اکسید آهن از 99/1 به 42/8 درصد و در بنتونیت سدیم از 43/2 به 43/0 درصد در بنتونیت پیلارد کاهش یافت. آفلاتوکسین باعث افزایش FCRو فعالیت آنزیمهای ALT و ALP در مقایسه با شاهد منفی شد. افزودن 25/0 درصد PB به جیرۀ آلوده باعث افزایش مصرف خوراک، وزن بدن و کاهش ALT شد. افزودن 75/0 درصد PB باعث کاهش ALT و ALP در مقایسه با تیمار آلوده شد. بنابراین بنتونیت سدیم پیلارد باعث بهبود عملکرد و کاهش فعالیت آنزیمهای کبدی در جوجههای تغذیهشده با آفلاتوکسین میشود. نتایج بهدستآمده در این بررسی، آفلاتوکسین در سطح 1000 نانوگرم در کیلوگرم اثری بر وزن بدن و مصرف خوراک نداشت و سطح 25/0 درصد بنتونیت پیلارد مناسبترین پاسخ عملکردی را در جوجههای تغذیهشده با آفلاتوکسین داشت.

بررسی فراسنجههای تولیدی، بیوشیمیایی، ایمنی و پاداکسندگی در مرغهای تخمگذار های-لاین W-36 تغذیهشده با جیرههای حاوی پادزیست محرک رشد و ترکیب افزودنیهای خوراکی
بهمنظور بررسی تأثیر برخی افزودنیهای خوراکی در مقایسه با پادزیست (آنتیبیوتیک) محرک رشد فلاوومایسین، آزمایشی در قالب طرح کامل تصادفی با سه تیمار، پنج تکرار و بیست قطعه مرغ در هر تکرار با استفاده از 300 قطعه مرغ تخمگذار لگهورن سفید، سویه های-لاین W-36 انجام شد. تیمارهای آزمایشی شامل شاهد، پادزیست محرک رشد فلاوومایسین به میزان 05/0 درصد جیره و ترکیب افزودنی خوراکی شامل عصارههای گیاهی آویشن و پونۀ کوهی، اسید پروپیونیک و مکمل روی به میزان 1/0 درصد جیره بود که در سه دورۀ متوالی دو هفتهای درمجموع به مدت 42 روز برای تغذیه در اختیار مرغهای تخمگذار قرار داده شدند. نتایج این بررسی نشان دادند، استفاده از ترکیب افزودنی خوراکی سبب بهبود معنیداری در ضریب تبدیل خوراک، درصد تولید تخممرغ و عیار پادتن (تیتر آنتیبادی) علیه آنفلوانزا و لیپوپروتئین با چگالی پایین در سرم خون نسبت به گروه شاهد شد (05/0≥P). مقادیر شاخصهای پاداکسندگی (آنتیاکسیدانی) سرم خون شامل غلظت سلنیوم، ویتامین E، سوپراکسید دیسموتاز و گلوتاتیون پراکسیداز در مرغهای تغذیهشده با جیرۀ حاوی ترکیب افزودنی خوراکی نسبت به دو گروه تیماری دیگر بهصورت معنیداری افزایش یافت (05/0≥P). تیمارهای آزمایشی تأثیر معنیداری بر صفات کیفی تخممرغ و فراسنجههای بیوشیمیایی خون به غیر از لیپوپروتئین با چگالی پایین نداشتند (05/0≤P). بنابر نتایج این آزمایش به نظر میرسد که افزودنی خوراکی مورد بررسی میتواند بهعنوان جایگزین پادزیست محرک رشد در جیرۀ غذایی مرغهای تخمگذار استفاده شود.

تأثیر مکمل بتائین و تزریق ویتامین B12 در دورۀ انتقال بر فراسنجههای سرم خون در گاوهای شیری
هدف از این تحقیق ارزیابی تأثیر مکمل بتائین و تزریق ویتامین B12 در دورۀ انتقال روی فراسنجههای سرم گاوهای شیری در دورۀ انتقال است. بدین منظور 32 گاو هلشتاین چند بار زایش کرده (21 روز پیشپیش از زایش) انتخاب شدند و به یکی از چهار تیمار آزمایشی تخصیص داده شدند: تیمار شاهد یا CC (بدون تزریق ویتامین B12 و دریافت مکمل بتائین)، تیمار دوم یا CB(تزریق هفتگی 10 سیسی ویتامین B12)، تیمار سوم یا BC (دریافت روزانه 50 گرم مکمل بتائین) و تیمار چهارم یا BB (تزریق هفتگی 10 سیسی ویتامینB12و دریافت روزانه 50 گرم مکمل بتائین). نمونههای خون در روزهای 7 روز پیش از زایش و همچنین 2 و 42 روز پس از زایش برای تعیین فراسنجههای خونی سرم گاوها و همچنین خون گوسالههای تازه متولدشده گرفته شد. فراسنجههای سرم خونی گاوها تحت تأثیر تیمارهای آزمایشی قرار نگرفتند (05/0P>)، بهجز برای گلوکز که در تیمارهای بتائین و B12 + بتائین نسبت به دو تیمار دیگر بهطور معنیداری بیشتر بود (05/0P<). در بین شاخصهای آنزیمی، گاماگلوتامیل ترانسفراز در تیمار B12 + بتائین در مقایسه با تیمار دوم (B12) کاهش معنیدار داشت (05/0P<). فراسنجههای سرمی گوسالههای تازه متولدشده از گاوهای دورۀ انتقال تحت تأثیر تیمارها قرار نگرفت. در نتیجه بتائین و ویتامین B12 تنها توانستند در بین فراسنجههای خونی گلوکز را افزایش دهند.

هدف از این بررسی ارزیابی تأثیر افزودن سطوح بالای آلفاتوکوفرول استات بر فراسنجههای بافتشناسی بیضۀ خروسهای سنگین مادر گوشتی بود. در این راستا، شمار 60 خروس مادر گوشتی راس 308 (با سن 20 هفته) در دو گروه وزنی استاندارد (30=شمار؛ kg169/0±109/3=میانگین وزن بدن) و سنگین (30=شمار؛ kg 161/0 ± 509/3= میانگین وزن بدن) تقسیم شدند. خروسها بهصورت تصادفی در ده گروه تیماری تقسیم و به قفسهای انفرادی منتقل شدند (بهصورت فاکتوریل 5×2 با شش تکرار در هر تیمار). تیمارهای آزمایشی با جیرهای همسان افزون بر سطوح مختلف ویتامین E 0 (E0)، ۱۰۰ (E100)، ۲۰۰ (E200)، ۳۰۰ (E300)، ۴۰۰ (E400) میلیگرم در کیلوگرم جیرۀ ویتامین E به مدت ده هفته تغذیه شدند. در مدت آزمایش، گروه سنگین 30 درصد بیشتر از گروه سبک خوراک دریافت کرد. در پایان آزمایش خروسها کشتار شدند و بیضهها برای ارزیابیهای بافتشناسی از لاشه جدا شدند. نتایج نشان داد، ویتامین E بهصورت معنیداری اندازۀ بیضهها و شمار لولههای اسپرمساز را در همۀ گروههای تیماری نسبت به گروه شاهد (کنترل) افزایش داده است (05/0P≤). همچنین، در هر دو گروه وزنی، قطر بیضۀ چپ، قطر لولههای اسپرمساز، ضخامت بافت پوششی (اپیتلیوم) اسپرمساز، شمار یاختههای لایدیگ و شمار رگهای خونی برای گروه 400 میلیگرم ویتامین E بالاتر بود (05/0P≤). درکل، بنابر نتایج بهدستآمده میتوان عنوان کرد، سطوح بالای ویتامین E (400 میلیگرم در کیلوگرم جیره) موجب بهبود فراسنجههای بافتشناسی بیضۀ خروس شده است.

تأثیر مکمل چربی (نمکهای کلسیمی) در جیرههای آغازین حاوی دو سطح پروتئین عبوری بر عملکرد، رشد و متابولیتهای خونی گوسالههای شیرخوار هلشتاین
اثر متقابل سطح چربی (0 و 30 گرم در کیلوگرم) و پروتئین عبوری (36 و 40 درصد پروتئین خام جیره) بر عملکرد، اندازههای بدنی (اسکلتی) و متابولیتهای خونی گوساله بررسی شد. از 40 رأس گوسالۀ شیرخوار نر هلشتاین با میانگین سن 39 روز در یک آرایش فاکتوریل با چهار تیمار استفاده شد. جیرههای آزمایشی شامل؛ بدون مکمل چربی با سطح پایین پروتئین عبوری (UF-LRUP)، با مکمل چربی و با سطح پایین پروتئین عبوری (SF-LRUP)، بدون مکمل چربی و با سطح بالای پروتئین عبوری (UF-HRUP)، با مکمل چربی و با سطح بالای پروتئین عبوری (SF-HRUP) بودند. نتایج نشان داد، استفاده از چربی سبب بهبود مصرف جیرۀ آغازین (استارتر)، افزایش وزن و همچنین بهبود بازدهی مصرف خوراک در گوسالههای شیرخوار شد (01/0>P). صفات عملکردی تحت تأثیر رابطۀ متقابل بین چربی و سطح پروتئین عبوری قرار نگرفت (05/0<P). از دیدگاه رشد، افزایش سطح چربی سبب افزایش طول بدن و عرض سینه شد و از سویی نیز سطح پروتئین عبوری بالاتر سبب افزایش ارتفاع کفل (هیپ) شد. عرض سینه و همچنین ارتفاع جدوگاه دامها نیز تحت تأثیر اثر متقابل چربی و سطح پروتئین عبوری قرار گرفت. با وجودی که سطح چربی مصرفی تأثیری بر فراسنجههای خونی نداشت، اما سطح بالاتر پروتئین عبوری سبب کاهش گلوکز خون و همچنین انسولین خون نیز شد (05/0>P). بهطورکلی، افزایش سطح چربی در جیرۀ گوسالههای شیرخوار پیش از شیرگیری قابل توصیه بوده و درصورتیکه مصرف آن با افزایش سطح پروتئین عبوری جیره باشد پاسخهای بهتری از نظر رشد به همراه خواهد داشت.

تأثیر مستندسازی نژادگان و معماریهای مختلف ژنگانی بر عملکرد روشهای جنگل تصادفی و بیز آستانهای A در پیشبینی ژنگانی
انتخاب ژنگانی (ژنومی) با بهرهگیری از مستندسازی (ایمپیوتیشن) میتواند نقش مهمی در افزایش بهرهوری اقتصادی و پیشرفت ژنتیکی صفات آستانهای ایفا کند. هدف این تحقیق بررسی درستی مستندسازی و تأثیر آن در سطح زیر منحنی مشخصۀ عملکرد (AUROC) ارزیابی ژنگانی روشهای بیز آستانهای A(TBA) و جنگل تصادفی (RF) در ویژگیهای آستانهای با معماریهای مختلف ژنگانی است. دادههای ژنگانی برای سطوح متفاوت وراثتپذیری (1/0 و 3/0)، سطوح مختلف LD (135/0 و 295/0) و شمار متفاوت جایگاههای ویژگیهای کمی (108 و 1080) روی کروموزم 27 کروموزم همانندسازی شدند. برای همانندسازی شرایط واقعی برای هر پیشفرض (سناریو)، از بین 54 هزار نشانگر همانندسازیشده بهطور تصادفی اقدام به حذف 50 درصد و 90 درصد نشانگرها کرده و در مرحلۀ بعد با مستندسازی اقدام به پیشبینی نژادگان (ژنوتیپ) نشانگرها کرده و درستی مستندسازی ارزیابی شد. در گام آخر، نژادگانهای اصلی و مستندشده با استفاده از روش TBA و RF برای ارزیابی AUROC استفاده شدند. با افزایش سطح LD و کاهش میزان حذف نشانگرها، درستی مستندسازی بهبود یافت. میانگین AUROC پیشفرضهای همانندسازیشده برای جنگل تصادفی و TBA بهترتیب 64/0 و 66/0 بود. استفاده از نژادگانهای مستندشده با میزان حذف 50 درصد و 90 درصد، بهترتیب AUROC را به میزان 013/0 و 02/0 برای RF و 018/0 و 026/0 برای TBA کاهش داد. بهرغم AUROC بالای روش بیز آستانهای A در پیشفرضهای مختلف، روش جنگل تصادفی عملکرد بهتری در شمار بالای QTL نشان داد. بهطورکلی استفاده از نژادگانهای مستندشده (k5) میتواند راهکار مهمی برای کاهش هزینههای ارزیابی ژنگانی باشد.

تأثیر جایگزینی بخشی از یونجه با تفالۀ چغندر، کاه بدون فرآوری و فراوریشده بر عملکرد گاوهای شیری پرتولید
در این پژوهش، تأثیر جایگزینی بخشی از یونجۀ جیرۀ غذایی با پسماندهای جانبی پر الیاف (تفاله و کاه) بر عملکرد گاوهای شیرده پرتولید بررسی شد. شمار هشت رأس ﮔﺎو هلشتاین پرتولید (5/2±54 کیلوگرم شیر) در قالب ﻃﺮح ﻣﺮﺑﻊ ﻻﺗﯿﻦ 4×4 به چهار جیرۀ آزمایشی اختصاص یافتند. جیرههای آزمایشی حاوی میزان ثابتی ذرت سیلویی (سیلاژ) (2/19%) با نسبت متفاوتی از 1) 2/19% یونجه، 2) 8/3% یونجه، 8/5% کاه گندم و 8/5% تفالۀ چغندر (کاه-تفاله)، 3) 8/3% یونجه و 5/8% کاه (کاه) و 4) 8/3% یونجه و 2/10% کاه فراوریشده بودند. سطح انرژی، پروتئین و الیاف (peNDF) در همۀ جیرهها یکسان بود، ولی الیاف هضمنشده (uNDF) در جیرههای حاوی تفاله-کاه و کاه فراوریشده کمتر بود. نتایج نشان داد، جایگزینی یونجه با کاه-تفاله یا کاه فراوریشده منجر به افزایش گوارشپذیری شد (01/0>P). همچنین، خوراک مصرفی در گاوهای تغذیهشده با منبعهای الیافی جایگزین بیشتر از گاوهای تغذیهشده با جیرۀ شاهد بود (01/0>P). تولید شیر و ترکیبهای شیر تحت تأثیر جیرههای آزمایشی قرار نگرفت. از همین رو، کارایی تولید شیر خام با تغذیۀ جیرۀ شاهد تمایل (08/0=P) به افزایش داشت. کارایی تولید شیر تصحیحشده برای چربی بین تیمارهای آزمایشی اختلافی نداشت. تغذیه با منبعهای الیافی مختلف تأثیری بر pHشکمبه، اسیدهای چرب فرّار و رفتار جویدن نداشت، اما نسبت استات:پروپیونات (06/0=P) را افزایش داد. جایگزینی بخشی از یونجه با کاه با/بدون تفاله منجر به افزایش گوارشپذیری و خوراک مصرفی شد ولی کارایی شیر خام تولیدی تمایل به کاهش داشت.
