مطالعه مقایسهای قابلیت هضم پیت نیشکر عملآوریشده با بخار آب تحت فشار توسط قارچهای بیهوازی شکمبه گاو هلشتاین و گاومیش خوزستان
این مطالعه به منظور مقایسة قابلیت هضم پیت نیشکر عملآوریشده توسط قارچها و کل میکروارگانیسمهای شکمبة گاو و گاومیش انجام شد. قابلیت هضم مادة خشک (DM)، الیاف نامحلول در شویندة خنثی (NDF) و الیاف نامحلول در شویندة اسیدی (ADF) پیت نیشکر عملآوریشده توسط کل میکروارگانیسمها و قارچهای شکمبة گاو و گاومیش به روش هضم دو مرحلهای، تکنیک تولید گاز و کشت اختصاصی قارچهای شکمبه اندازهگیری و مقایسه شد. قابلیت هضم آزمایشگاهی مادة خشک، NDF و ADF پیت نیشکر توسط کل میکروارگانیسمهای شکمبة گاومیش (به ترتیب 62، 31/32 و 22 درصد) بیشتر از گاو (به ترتیب 13/50، 07/27 و 25/16 درصد) بود (05/0P<). صرف نظر از نوع میکروارگانیسم قابلیت هضم مادة خشک NDF و ADF توسط گاومیش (13/54، 51/27 و 86/19 درصد) بیشتر از گاو (69/49، 54/24 و 67/14 درصد) بود (05/0P<). پتانسیل تولید گاز پیت نیشکر عملآوریشده در حضور کل میکروارگانیسمهای مایع شکمبة گاو از نظر عددی بیشتر از گاومیش بود (05/0P>). نرخ تولید گاز توسط کل میکروارگانیسمها و قارچهای شکمبة گاومیش به طور معناداری بیشتر از گاو بود (05/0P<). صرف نظر از نوع میکروارگانیسم، نرخ تولید گاز پیت نیشکر در گاومیش به طور معناداری بیشتر از گاو بود (05/0P<)؛ برعکس پتانسیل تولید گاز در گاو اندکی بیشتر بود (05/0P>). صرف نظر از نوع دام، قابلیت هضم و توان تولید گاز برای کل میکروارگانیسمها بیشتر از قارچها بود (05/0P<)، اما برای نرخ تولید گاز بین آنها تفاوتی وجود نداشت. در محیط کشت اختصاصی قارچهای شکمبه، قابلیت هضم مادة خشک پیت نیشکر توسط قارچها در گاومیش در روز دوازدهم بهطور معناداری بیشتر از گاو بود (05/0P<). تراکم قارچها در هر میلیلیتر مایع شکمبة گاو بیشتر از گاومیش بود (05/0P<). در کل، با وجود تعداد بیشتر قارچهای شکمبة گاو، میتوان گفت که توان قارچها و کل میکروارگانیسمهای شکمبة گاومیش در آزمایش حاضر بیشتر از گاو بود. بنابراین، نتایج برتری گاومیش به گاو هلشتاین در استفاده از مواد فیبری کمکیفیت را نشان داد.

مطالعة تأثیرات استفاده از جلبک گراسیلاریوپسیز پرسیکا در جیرة مرغان تخمگذار بر عملکرد، صفات کیفی تخممرغ و لیپیدهای سرم
این آزمایش به منظور بررسی تأثیرات استفاده از جلبک گراسیلاریوپسیز پرسیکا در جیرة غذایی مرغهای تخمگذار بر عملکرد، صفات کیفی تخممرغ و لیپیدهای سرم اجرا شد. آزمایش در قالب طرح کاملاً تصادفی با پنج تیمار شامل جیرههای حاوی سطوح صفر، 5، 10، 15 و 20 درصد جلبک، در پنج تکرار و با چهار قطعه مرغ در هر تکرار و در مجموع 100 قطعه مرغ سویههای لاین w-36، به مدت 12 هفته (23 تا 34 هفتگی) انجام گرفت. استفاده از جلبک در سطوح 10 درصد و بیشتر موجب کاهش تخمگذاری و افزایش ضریب تبدیل در مقایسه با گروه شاهد شد (05/0>P). تودة تخممرغ در پرندگانی که جیرة حاوی سطوح 15 و 20 درصد جلبک را دریافت کردند، کمتر از تیمار شاهد بود (05/0>P). ارتفاع سفیدة تخممرغ در پرندگان تغذیهشده با سطوح بیش از 5 درصد جلبک در جیره، کمتر از تیمار شاهد بود (05/0>P). استفاده از سطوح مختلف جلبک در جیره، ضخامت پوسته و وزن مخصوص تخممرغ را بهطور معناداری در مقایسه با گروه شاهد افزایش داد (05/0>P). زردة تخمهای مرغهایی که جیرههای حاوی جلبک دریافت کردند، مالون دی آلدئید کمتر و ید بیشتری در مقایسه با شاهد داشت (05/0>P). استفاده از جلبک در جیره بر مقدار کلسترول زردة تخممرغ تأثیری نداشت، ولی سطوح 15 و 20 درصد جلبک در جیره، کلسترول سرم را کاهش داد (05/0>P). بر اساس نتایج، میتوان از جلبک گراسیلاریوپسیز پرسیکا تا سطح 5 درصد بدون تأثیر منفی بر عملکرد در جیرة مرغهای تخمگذار استفاده کرد و علاوه بر بهبود ضخامت پوسته و چگالی تخممرغ، محتوای ید در تخممرغ و همچنین مدت زمان انبارداری آن را افزایش داد.

مطالعة آناتومی سرویکس گوسفند و تأثیر آن بر مقدار نفوذ گان تلقیح به درون سرویکس
یکی از محدودیتها در تلقیح مصنوعی گوسفند ساختار ویژة سرویکس گوسفند است که دارای تفاوتهای چشمگیری است. موفقیت در تلقیح مصنوعی با مورفولوژی سرویکس گوسفند ارتباط تنگاتنگی دارد. هدف از این پژوهش مطالعة ساختار آناتومی سرویکس و مقایسة آن در گوسفندهای نژاد ماکویی، کردی و افشاری است. در این پژوهش 300 دستگاه تولید مثلی گوسفند ماده از نژادهای یادشده (از هر نژاد 100 رأس) در فصل تولید مثل از کشتارگاه استان تهران و استان البرز جمعآوری شد. شکل خارجی، دهانه، طول و قطر سرویکس، مقدار نفوذ، نوع و تعداد رینگ بررسی گردید. بیشترین فراوانی در شکل خارجی دهانة نوک پستانی و سرویکس با طول 3 تا 4 سانتیمتر مشاهده شد. شکل غالب دهانة سرویکس در نژادهای کردی و افشاری نوک پستانی بود، ولی در گوسفند ماکویی شکل فلپ غالب است. نتایج نشان داد میانگین قطر سرویکس برای کل دادهها 7/1±34/9 میلیمتر است. میانگین تعداد رینگ در بین نژادهای مورد مطالعه 8/0±59/5 عدد بود. بیشترین فراوانی آرایش و طرز قرارگرفتن رینگها در کانال سرویکس، آرایش دستة دوم را نشان داد. همبستگی طول سرویکس با تعداد رینگ 24/0 (001/0p<) و همبستگی طول با قطر سرویکس 25/0 (001/0p<) محاسبه شد. طول سرویکس با زاویة رینگ، شکل دهانة سرویکس و سن همبستگی معناداری نشان نداد (05/0<p). طول سرویکس، قطر و تعداد رینگ، در بین نژادهای مورد بررسی تفاوت معناداری نشان نداد (05/0<p). میانگین نفوذ گان تلقیح به درون کانال سرویکس برای کل دادههای سرویکس 5/2±92/3 میلیمتر و به طور میانگین برابر با 2/2±24/4، 1/1±46/3 و 5/2±84/3 برای نژادهای ماکویی، کردی و افشاری بود. مقدار نفوذ گان تلقیح به سرویکس، تحت تأثیر قطر سرویکس قرار گرفت (001/0p<). چرخة فحلی بر عمق نفوذ تأثیر معناداری داشت، به طوری که عمق نفوذ گان تلقیح در گامة لوتیال به درون سرویکس کمتر و در زمانی که تخمدان فاقد جسم زرد باشد بیشتر است. مقدار نفوذ گان تلقیح تحت تأثیر تعداد رینگ قرار داشت، به طوری که سرویکسهای با تعداد رینگ بیشتر، نفوذ کمتری داشتند. فراسنجههای بررسیشده، به جز شکل خارجی دهانة سرویکس در بین سه نژاد مورد مطالعه تفاوتی نداشت.

تعیین تراکم و ضرایب قابلیت هضم حقیقی اسیدهای آمینة پودر گوشت و استخوان در خروسهای بالغ لگهورن سکومبرداریشده
تراکم اسیدهای آمینة 6 نمونه پودر گوشت و استخوان به روش کروماتوگرافی تبادل یونی و قابلیت هضم حقیقی اسیدهای آمینه به روش تغذیة دقیق سیبالد تعیین شد. متیونین با 35/0 و لوسین با 29/2 درصد کمترین و بیشترین مقدار اسیدهای آمینه را در بین نمونههای پودر گوشت و استخوان نشان دادند. میانگین مقادیر هریک از اسیدهای آمینه، مجموع اسیدهای آمینة ضروری و غیرضروری و کل اسیدهای آمینه از مقادیر مشابه در جدولهای NRC (1994) کمتر بود. نتایج نشان داد که به جز متیونین و پرولین، ضرایب قابلیت هضم حقیقی هر یک از اسیدهای آمینه، کل اسیدهای آمینة ضروری، غیرضروری، اسیدهای آمینة گوگرددار و کل اسیدهای آمینه تفاوت معناداری بین نمونههای پودر گوشت و استخوان نشان ندادند. میانگین ضریب قابلیت هضم حقیقی اسید آمینة سیستئین و آرژنین معادل 7/73 و 4/89 درصد بود که به ترتیب کمترین و بیشترین مقدار را در بین اسیدهای آمینة پودر گوشت و استخوان نشان داد. گرچه ضرایب قابلیت هضم اسیدهای آمینة پودر گوشت و استخوان در سطح بالایی قرار داشت، اما به دلیل کمبودن تراکم اسیدهای آمینه، از سطح اسیدهای آمینة قابل هضم کمتری برخوردار بود.

اثر افزودن سطوح متفاوت سولفات روی به رقیقکنندة منی بر کیفیت اسپرم گاو پس از انجماد-ذوب
هدف از این پژوهش، مقایسة اثر افزودن سطوح متفاوت روی به رقیقکنندة منی بر ویژگیهای حیاتی اسپرم پس از انجماد در گاو بود. اسپرمگیری از چهار گاو نر هلشتاین هفتهای دو بار انجام شد و سپس نمونههای مناسب با هم مخلوط شدند. نمونههای اسپرم (در پنج تکرار) به چهار گروه شامل سطوح صفر، 50، 100 و 150 میکرومول روی (Zn-0, Zn-50-Zn-100, Zn-150) اختصاص داده و منجمد شدند. نمونههای اسپرم بعد از انجماد از نظر جنبایی و ریختشناختی، زندهمانی، یکپارچگی غشا، سلامت آکروزوم، پراکندگی کروماتین اسپرم و تولید مالوندیالدئید بررسی شدند. اختلاف معناداری در درصد جنبایی و ریختشناسی اسپرم در محیطهای دارای روی با گروه شاهد وجود نداشت. گروههای Zn-100 و Zn-150 دارای درصد بیشتری از اسپرمهای با آکروزوم سالم در مقایسه با شاهد بودند (005/0 p<). زندهمانی و یکپارچگی غشا در گروه Zn-100 از گروه شاهد بهتر بود (05/0P< و 01/0 P<). درصد اسپرمهای با DNA آسیبدیده در گروه Zn-150 در گروه مقایسه با گروههای دیگر بیشتر بود (005/0 p<). درصد تولید مالوندیآلدهید در گروه Zn-100 و Zn-150 از بقیة گروهها کمتر بود (05/0 P<). بنابراین استفاده از روی در رقیقکنندة منی گاو سبب بهبود برخی فراسنجههای اسپرم بعد از فرایند انجماد میشود.

تأثیر تغذیة پیوگلیتازون بر تخمکریزی و برخی بیماریهای پس از زایش در گاوهای هولشتن
هدف این پژوهش بررسی اثر پیوگلیتازون بر کنش تخمدان و کاهش برخی بیماریهای پس از زایش در گاوهای هولشتن بود. تعداد هشتاد گاو شیری هولشتن بهصورت تصادفی در چهار گروه تیماری (20n=) با آرایش فاکتوریل 2×2 قرار گرفتند. تغذیة پیوگلیتازون از 14 روز مانده به زمان مورد انتظار زایش تا 21 روز پس از زایش انجام شد. تغذیة پیوگلیتازون پیش و پس از زایش، عامل اصلی در نظر گرفته شد؛ گاوها در هر دوره با یکی از دو سطح صفر یا 6 میلیگرم به ازای هر کیلوگرم وزن بدن پیوگلیتازون تغذیه شدند. فراسنجههای وابسته به سلامت گاو و روز تخمکریزی رکوردبرداری و دادهها با نرمافزار SAS و رویههای GENMOD یاGLM واکاوی شدند. تغذیة پیوگلیتازون در دورة پس از زایش، میانگین فاصلة زایش تا نخستین تخمکریزی را کاهش داد (02/0P<). افزون بر این، شمار گاوهایی که در 14 روز نخست پس از زایش تخمکریزی کردند نیز با تغذیة پیوگلیتازون در دورة پس از زایش افزایش یافت (03/0P<). تغذیة پیوگلیتازون در دورههای پیش از زایش (01/0P<) و پس از زایش (10/0P<) بسامد بروز لنگش را کاهش داد. پیوگلیتازون اثر معناداری بر بسامد ورم پستان نداشت. تغذیة پیوگلیتازون در دورههای پیش از زایش (03/0P<) و پس از زایش (08/0P<) درصد گاوهای دچار عفونتهای رحم را کاهش داد. تغذیة پیوگلیتازون پس از زایش،کیستهای تخمدانی را کاهش داد (04/0P<). درصد گاوهایی که تخمدان نافعال داشتند تحت تأثیر پیوگلیتازون قرار نگرفت. یافتهها نشان دادند که تغذیة پیوگلیتازون به گاوهای شیری دردورة انتقال میتواند سبب بهبود کنشهای تخمدان شود و در عین حال برخی بیماریهای پس از زایش را کاهش دهد.

بررسی تأثیر پودر و اسانس آویشن بر پارامترهای عملکرد، کیفیت گوشت و رودة بلدرچینهای گوشتی
به منظور بررسی تأثیر پودر و اسانس آویشن بر عملکرد بلدرچینهای گوشتی در مقایسه با آنتیبیوتیک، آزمایشی در قالب طرح کاملاً تصادفی با 6 تیمار و 4 تکرار به اجرا درآمد. تیمارهای آزمایشی شامل جیرة پایه بدون افزودنی خوراکی یا با 100 میلیگرم در کیلوگرم آنتیبیوتیک ویرجینیامایسین و سطوح مختلف 1/0 و 2/0 درصد پودر و 100 و 200 میلیگرم در کیلوگرم اسانس آویشن بودند. پارامترهای اندازهگیریشده شامل عملکرد، کیفیت گوشت، ریختشناسی و میکروبیولوژی روده بودند. در 35-0 روزگی افزودن 1/0 درصد پودر آویشن و آنتیبیوتیک سبب افزایش وزن بدن شد. پودر و اسانس آویشن تیوباربیتوریک اسید، اُفت خونابه[1] و اُفت در نتیجة پخت[2] را کاهش و ظرفیت نگهداری آب را افزایش داد. بیشترین تعداد باکتری لاکتوباسیل در مقایسه با گروه شاهد، در گروههای تغذیهشده با آویشن (به جز 200 میلیگرم در کیلوگرم اسانس) و کمترین تعداد در تیمار دریافتکنندة آنتیبیوتیک مشاهده شد. کمترین تعداد باکتریهای اشریشیاکلی مربوط به تیمار دریافتکنندة آنتیبیوتیک بود. بیشترین طول و عرض پرز و کمترین عمق کریپت در گروه تغذیهشده با آویشن (به جز 200 میلیگرم در کیلوگرم اسانس) مشاهده شد. در گروه آنتیبیوتیک علاوه بر طول و عرض پرزها، عمق کریپت نیز افزایش معناداری پیدا کرد. تأثیر سه نوع برگ گیاه اکالیپتوس بر تخمیرشکمبهای، جمعیت پروتوزوآیی و تولید گاز متان به روش برونتنی
در این مطالعه تأثیر سه سطح برگ اکالیپتوس (0، 65/28 و 31/57 میلیگرم) که به سه روش هوا، آون و برودت-خشک، خشک شده بودند، بر خصوصیات تخمیر شکمبة گوسفند به روش برونتنی و آزمون گاز بررسی شد. مقادیر گاز کل تولیدی و متان تولیدی، نیتروژن آمونیاکی، تجزیهپذیری آزمایشگاهی مادة آلی، شاخص بازده سنتز پروتئین میکروبی (PF) (میلیگرم مادة آلی تجزیهشده بخش بر میلیلیتر گاز تولیدی) و غلظت اسیدهای چرب فرار فراسنجههایی بود که مطالعه شد. همچنین سه زیرخانوادة Entodiniinae، Ophryscolecinae، Diplodiniinae و یک خانوادة Isotrichdae و جمعیت پروتوزوآیی کل شناسایی و شمارش شدند. برگ اکالیپتوس تهیه شده به روشهای هوا - خشک (خطی 01/0>P) و برودت-خشک (غیرخطی 01/0>P) مقدار اسیدهای چرب فرار کل را کاهش داند. اما، تنها روش آون-خشک، اسید پروپیونیک را افزایش (خطی 05/0>P) داد. افزودن برگ اکالیپتوس، فراسنجههای گاز تولیدی از بخش نامحلول (01/0>P، خطی)، گاز تولیدی در زمانهای 24 و 54 ساعت (01/0>P، خطی و غیرخطی)، گاز متانتولیدی (05/0>P، خطی)، نیتروژن آمونیاکی (01/0>P، خطی) و اسیدهای چرب فرار کل را کاهش (01/0>P، خطی) داد. اما شاخص PF تنها درسطح 31/57 برگ اکالیپتوس بهبود یافت (01/0>P، خطی). همچنین، این گیاه دارای فعالیت ضد پروتوزوآیی بوده و سبب کاهش جمعیت پروتوزوآیی کل، زیر خانوادة Entodiniinae (01/0>P، خطی) و خانوادة Isotrichdae (01/0>P، خطی) شد. نتایج این تحقیق نشان میدهد که برگ اکالیپتوس توانایی بهبود تخمیر شکمبهای در شرایط برونتنی را دارد و روش آون- خشک مؤثرتر از دو روش دیگر بوده است.

بررسی تأثیر کودهای نیتروژن و فسفر بر ارزش غذایی علوفه و سیلاژ گلرنگ
تحقیق حاضر به منظور بررسی اثر سیلوکردن و کاربرد کودهای نیتروژن و فسفربر ترکیب شیمیایی و فراسنجههای تجریهپذیری شکمبهای مادة خشک گیاه گلرنگ Carthamustinctorius L.)) انجام گرفت. گیاه گلرنگ در آذرماه 1388 کشت گردید و در اواخر خرداد 1389 در مرحلة گلدهی برداشت شد، با استفاده از دستگاه خردکن به قطعات 5-3 سانتیمتری تبدیل و بخشی از آن در سیلوهای آزمایشی برای مدت 60 روز ذخیره شد. تیمارهای سیلویی آزمایشی عبارت بود از: علوفة کوددهی نشده، علوفة کوددهی شده با فسفر (100 کیلوگرم در هکتار)، علوفة کوددهی شده با نیتروژن (300 کیلوگرم در هکتار)، علوفة کوددهی شده با نیتروژن و فسفر (300 کیلوگرم نیتروژن و 100 کیلوگرم فسفر در هکتار)، سیلاژ حاصل از علوفة کوددهی نشده، سیلاژ حاصل از علوفة کوددهی شده با فسفر (100 کیلوگرم در هکتار)، سیلاژ حاصل از علوفة کوددهی شده با نیتروژن (300 کیلوگرم در هکتار) و سیلاژ حاصل از علوفة کوددهی شده با نیتروژن و فسفر (300 کیلوگرم نیتروژن و 100 کیلوگرم فسفر در هکتار). ترکیب شیمیایی، تجزیهپذیری مادة خشک و قابلیت هضم با استفاده از روشهای استاندارد برآورد گردید. کوددهی با نیتروژن و فسفر سبب کاهش مقدار الیاف نامحلول در شویندة خنثی و اسیدی و افزایش مقدار خاکستر، کلسیم و فسفر گیاه شد (05/0>P). کوددهی با نیتروژن سبب افزایش مقدار پروتئین خام، نیتروژن غیرپروتئینی، پروتئین محلول در بافر فسفات بورات و کاهش نیتروژن نامحلول در شویندة اسیدی گیاه شد (05/0>P). کوددهی با نیتروژن و فسفر و سیلوکردن گیاه سبب افزایش بخش سریع تجزیة مادة خشک، ثابت نرخ تجزیهپذیری و تجزیهپذیری مؤثر شکمبهای مادة خشک شد (05/0>P). همچنین کوددهی با نیتروژن و فسفر و نیز سیلوکردن گیاه موجب افزایش گاز تولیدی حاصل از بخش نامحلول، ثابت نرخ تولید گاز، انرژی قابل متابولیسم و مادة آلی قابل هضم آن شد. در مجموع استفاده از سیلاژ علوفة گلرنگ کوددهی شده با نیتروژن و فسفر در مقایسه با علوفة کوددهی نشده و سیلونشده توصیه میشود.

اثر سطوح مختلف کود مرغی عملآوریشده بر مصرف خوراک، قابلیت هضم، عملکرد و متابولیتهای شکمبه و خون برههای نر مغانی
این پژوهش در قالب طرح کاملاً تصادفی با چهار تیمار و نه تکرار روی 36 رأس برة نر مغانی انجام گرفت که در آن چهار جیرة غذایی حاوی سطوح صفر (شاهد)، 7، 14 و 21 درصد کود مرغی فراوریشده، آزمایش شد. نتایج نشان داد که مصرف سطوح مختلف کود مرغی درجیره، بر مصرف اختیاری خوراک و افزایش وزن برهها در طول دورة آزمایش اثری نداشت (05/0< P). با افزایش کود مرغی در جیره قابلیت هضم مادة خشک و NDF بهطور خطی کاهش یافت (05/0P<)، ولی قابلیت هضم مادة آلی و پروتئین خام تحت تأثیر جیرههای آزمایشی قرار نگرفت (05/0< P). با گنجاندن کود مرغی در جیرة غذایی، غلظت استات در شکمبه به طور خطی کاهش یافت، اما غلظت آمونیاک افزایش نشان داد (05/0P<)، در حالی که غلظت پروپیونات، بوتیرات، والرات، ایزووالرات ونسبت استات به پروپیونات تحت تأثیر جیرههای آزمایشی قرار نگرفت (05/0< P). نیتروژن اورهای خون با افزایش کود مرغی در جیره بهطور خطی افزایش یافت، اما غلظت سایر متابولیتهای خون تحت تأثیر جیرههای آزمایشی قرار نگرفت (05/0< P). در مجموع، مصرف کود مرغی در جیرة برههای مغانی تا سطح 21 درصد، بر مصرف مواد مغذی و عملکرد رشد اثری نداشت ولی قابلیت هضم مادة خشک و NDF و غلظت استات شکمبه را کاهش داد. با افزایش سطح کود مرغی در جیره، هزینة خوراک به ازای هر واحد افزایش وزن زندة برهها، بهطور خطی کاهش یافت (05/0P<). بهطور کلی میتوان نتیجهگیری کرد که مصرف کود مرغی عملآوریشده تا 21 درصد جیرة غذایی برة پرواری امکانپذیر است.
