اثر سطوح مختلف گلوتاتیون درونپوشانیشده بر کیفیت انجماد اسپرم گاو
هدف از این مطالعه، ارزیابی اثر سطوح مختلف گلوتاتیون احیا در رقیقکننده بر کیفیت اسپرم گاو پس از فرایند انجماد و ذوب بود. در پژوهش حاضر، رقیقکننده حاوی نانولیپوزومهای غنیشده با چهار سطح صفر، 1، 5/2 و 5 میلیمولار آنتی اکسیدان گلوتاتیون بود. لیپوزومهای حاوی گلوتاتیون با استفاده از روش فیلم نازک تهیه شد. اندازه ذرات با استفاده از دستگاه سونیکاتور به ابعاد نانو کاهش یافت. چهل انزال طی شش هفته از شش راس گاو نر هلشتاین جمعآوری و مورد انجماد قرار گرفت. صفات مورد ارزیابی پس از انجماد و ذوب شامل، فراسنجههای جنبایی، فعالیت غشا و یکپارچگی غشا و ریختشناختی اسپرم بود. نتایج این مطالعه نشان داد که استفاده از 1 میلیمولار گلوتاتیون جنبایی کل و جنبایی پیشرونده را بهطور معنیداری افزایش میدهد (7/1±5/47 و 2±7/32 درصد)، اگرچه، تفاوت معنیداری با غلظت 5/2 میلیمولار نداشت (7/1±0/45 و 2±2/26 درصد). همچنین، غلظت 5/2 میلیمولار بالاترین میزان یکپارچگی و سلامت غشا را نشان داد، اما تفاوت معنیداری با غلظت 1 میلیمولار نشان نداد. با اینحال، 5 میلیمولار گلوتاتیون درون پوشانی شده سبب کاهش فراسنجههای کیفی اسپرم شد. بنابراین، استفاده از سطح 1 میلیمولار گلوتاتیون به شکل درونپوشانی شده در رقیقکننده میتواند حفاظت بهتری از اسپرم گاو نسبت به سایر سطوح به عمل آورد.

اثر منابع مختلف روی، بر عملکرد و آنزیمهای کبد جوجههای گوشتی
برای مقایسه آثار منابع آلی، غیرآلی و هیدروکسی روی بر عملکرد رشد و آنزیمهای کبد جوجه گوشتی، آزمایشی با استفاده از 360 قطعه جوجه راس 308 در قالب طرح کاملاً تصادفی با 6 تیمار و 4 تکرار انجام شد. تیمارهای آزمایشی عبارت بودند از 1) شاهد منفی؛ بدون مکمل روی، 2) شاهد مثبت دارای 80 میلیگرم سولفات روی در کیلوگرم جیره، 3) جیره دارای 80 میلیگرم هیدروکسیکلراید روی در کیلوگرم جیره، 4) جیره دارای 60 میلیگرم سولفات روی+ 20 میلیگرم روی-متیونین در کیلوگرم جیره، 5) جیره دارای 40 میلیگرم سولفات روی+ 40 میلیگرم اکسید روی در کیلوگرم جیره، 6) جیره دارای 30 میلیگرم سولفات روی + 30 میلیگرم اکسید روی + 20 میلیگرم روی-متیونین در کیلوگرم جیره. در پایان آزمایش، وزن نسبی بورس فابریسیوس و تیموس در مقایسه با گروه شاهد منفی کاهش یافت (05/0P<). منبع هیدروکسیکلراید روی مصرف خوراک را نسبت به گروه شاهد مثبت و افزایش وزن روزانه را در مقایسه با تیمارهای 1، 2 و 4 بهبود بخشید (05/0P<). فعالیت آلکالین فسفاتاز پلاسما در تیمارهای 5 و 6 نسبت به تیمارهای 3 و 4 افزایش معنیداری نشان داد، ولی تفاوت معنیداری در سطح لاکتاتدهیدروژناز، آسپارتاتآمینوترانسفراز و آلانین آمینوترانسفراز پلاسما در تیمارهای مختلف مشاهده نشد. یافتهها نشان میدهد منبع هیدروکسیکلراید روی آثار قابل مقایسهای با منابع غیرآلی و ترکیب منابع آلی و غیرآلی روی در جوجه گوشتی دارد.

شناسایی تنوع تعداد کپی و تأثیرات آنها بر ژنهای شترهای تککوهانه ایرانی با استفاده از دادههای توالییابی کامل ژنوم
مطالعه حاضر بهمنظور شناسایی تنوع تعداد کپی و بررسی اثرات آنها بر ژنهای شترهای تککوهانه با استفاده از دادههای توالییابی کامل ژنومی دو نفر شتر ایرانی (شتر یزدی و شتر طرودی) انجام شد. توالییابی کامل ژنوم نمونههای یزدی و طرودی، به ترتیب حدود 456 و 8/418 میلیون خوانش با اندازه 100 جفتبازی تولید کرد. پس از همردیفی خوانشهای پالایش شده در ژنوم مرجع (شماره دسترسی در NCBI: GCA_000767585.1)، از الگوریتم مبتنی بر عمق خوانش، برای شناسایی تنوع تعداد کپیها استفاده شد. تنوع تعداد کپی شناساییشده برای نمونههای موردمطالعه برابر با 831 برای شتر یزدی و 312 برای شتر طرودی بود. نزدیک به 60 درصد (606 ژن برای شتر یزدی و 288 ژن برای شتر طرودی) از تنوعات شناساییشده، با ژنها دارای تداخل بودند و ما بقی در نواحی خارج ژنی قرار داشتند. نتایج بهدستآمده نشان داد که ژنهای مهمی از جمله ژنهای دخیل در عملکرد سیستم ایمنی و مرگ سلولی برنامهریزیشده دارای تنوع تعداد کپی هستند. همچنین دو ژن مهم OOEP و WWC3 که در عملکرد تولیدمثلی پستانداران نقش دارند، در نمونههای مورد مطالعه دارای تنوع تعداد کپی بودند.

بررسی بیان ژن فاکتور رشد شبهانسولین-1 (IGF-I) و تغییرات بافتشناسی بیضهها در دوره بلوغ خروسهای بومی
بهمنظور مطالعه تغییرات رشد بیضهها و بیان ژن IGF-I در دوره بلوغ خروسهای بومی فارس، 24 قطعه خروس پنج، شش، هفت و هشت ماهه بهطور تصادفی انتخاب شدند. در هر نوبت نمونهبرداری شش خروس کشتار و نمونههای بافت بیضه برای اندازهگیری میزان فراوانی رونوشت ژن IGF-I و صفات بیومتریک بیضهها جمعآوری شدند. نتایج نشان داد که بیان ژن IGF-I در بیضه خروسهای شش، هفت و هشت ماهه نسبت به خروسهای پنج ماهه افزایش یافت، درحالیکه فراوانی رونوشت این ژن در هفت ماهگی تفاوتی با شش ماهگی نداشت. همچنین، نتایج نشان دادند که وزن بیضههای چپ و راست در هفت و هشت ماهگی نسبت به پنج و شش ماهگی افزایش یافت و بیشینه نسبت طول به عرض بیضه چپ در شش ماهگی و کمینه آن در هفت و هشت ماهگی مشاهده شد ولی این نسبت در بیضه راست در سنین مختلف تفاوتی نداشت. بررسی تغییرات بافتشناسی بیضهها نیز نشان داد که بیشینهی تعداد سلول های سرتولی و لایدیگ در هر دو بیضه چپ و راست مربوط به سن هشت ماهگی بود. بهطور کلی، نتایج نشان داد که فراوانی رونوشت ژن IGF-I در دوره بلوغ خروسهای بومی فارس (پنج تا هشت ماهگی) روندی افزایشی داشت و وزن بیضههای چپ و راست در هفت و هشت ماهگی به بیشترین مقدار خود رسید.

آثار استفاده از دو سطح اکسید روی با اندازه ذرات متفاوت بر عملکرد تولید مثلی مرغها و خروسهای مادر گوشتی
در تحقیق حاضر اثر سطوح و اندازه ذرات مختلف اکسید روی بر عملکرد تولید مثلی مرغها و خروسهای مادر گوشتی مورد ارزیابی قرار گرفت. از تعداد 200 قطعه مرغ مادر گوشتی سویه راس 308 به صورت طرح کاملاً تصادفی با 4 تیمار، 5 تکرار و 10 پرنده در هر تکرار و 24 قطعه خروس مادر گوشتی سویه راس 308 در قالب طرح کاملاً تصادفی با چهار تیمار، شش تکرار و یک پرنده در هر تکرار در سن 54 هفتگی استفاده شد. تیمارهای آزمایشی شامل جیره حاوی 70 میلیگرم روی از منبع اکسید روی با اندازه ذرات بزرگ(LPZnO-70)، 100 میلیگرم روی از منبع اکسید روی با اندازه ذرات بزرگ (LPZnO-100)، 70 میلیگرم روی از منبع اکسید روی با اندازه ذرات کوچک (SPZnO-70) و 100 میلیگرم روی از منبع اکسید روی با اندازه ذرات کوچک (SPZnO-100) در هر کیلوگرم خوراک بودند. نتایج نشان دادند که تیمارهای آزمایشی بر تولید تخممرغ، وزن بدن، وزن تخممرغ و وزن زرده تخممرغ تاثیر معنیداری نداشتند. ضخامت پوسته تخممرغ در کل دوره آزمایش در تیمار حاوی SPZnOبیشتر از LPZnO بود (001/0P<). بالاترین و پایینترین میزان باروری و جوجهدرآوری بهترتیب در تیمارهای حاوی SPZnO-100وLPZnO-70مشاهده شد. بیشترین میزان حجم مایع منی در تیمارهای حاوی SPZnO-100 و LPZnO -100 مشاهده شد (05/0P<). بهطور کلی میتوان نتیجه گرفت که SPZnO به دلیل قابلیت دسترسی بالاتر، تأثیر مطلوبتری بر فراسنجههای تولیدمثلی گلههای مادر گوشتی دارد. از آنجایی که سطح 100 میلیگرم روی در هر کیلوگرم جیره از هر دو منبع اکسید روی عملکرد تولیدمثلی بهتری را به همراه داشت، استفاده از SPZnO -100 توصیه میشود.

تغذیه گیاهان شورزیست به برههای پرواری و تأثیر آنها بر عملکرد، قابلیت هضم، فراسنجههای خونی و خصوصیات لاشه
بهمنظور بررسی تأثیر تغذیه گیاهان شورزیست گتک (Halocnemum strobilaceum) و سیاه شور مصری (aegyptiacaSuaeda) بر عملکرد، قابلیت هضم، فراسنجههای خونی و خصوصیات لاشه برههای تغذیهشده با جیرههای پرکنسانتره سی و شش رأس بره نر نژاد عربی با میانگین سنی 2/0±5/4 ماه و وزن 13/1±3/25 کیلوگرم بهطور تصادفی بهمدت 80 روز با جیرههای آزمایشی تغذیه شدند. جیرهها شامل 1- شاهد، 2- دارای 15 درصد گیاه سیاه شور مصری، 3- دارای 15 درصد گیاه گتک و 4- دارای 15 درصد گیاه سیاه شور مصری و 15 درصد گیاه گتک بود. تیمارها تفاوتی از نظر میانگین ماده خشک مصرفی، افزایش وزن روزانه، ضریب تبدیل غذایی و افزایش وزن نهایی نداشتند. مصرف علوفه شورزیست میزان آب مصرفی را افزایش داد (001/0p <). قابلیت هضم چربیخام (001/0p <) و الیاف نامحلول در شوینده اسیدی در تیمارهای شاهد و تیمار حاوی 15 درصد علوفه گیاه سیاه شور بالاتر از دیگر تیمارها بود (003/0P=). ماده آلی در تیمارهای حاوی 15 درصد گیاه سیاه شور و دارای 15 درصد علوفه گیاه گتک نسبت به تیمارهای دیگر قابلیت هضم بیشتری داشت. غلظت نیتروژن اورهای خون در پلاسمای برههای تیمار 30 درصد گیاه شور نسبت به برههای دیگر کمتر بود (0001/0P=). لاشه برههایی که30 درصد گیاه شور مصرف کردند، چربی کمتر (024/0P=) و گوشت بیشتری داشت (006/0P=). برههای تغذیه شده با 15 درصد سیاه شور، عضله راسته روشنتری داشتند (03/0P=). در کل، نتایج آزمایش نشان داد که استفاده از گیاهان شورزی سیاه شور و گتک جایگزین مخلوط کاه و یونجه در جیره عملکرد پرواری دام را حفظ نموده و چربی لاشه را کاهش میدهد.

اثر اندازه ذرات ذرت و منابع مختلف فیبر نامحلول بر عملکرد و خصوصیات لاشه و دستگاه گوارش جوجههای گوشتی
این پژوهش بهمنظور مطالعه اثر اندازه ذرات ذرت و منابع مختلف فیبر نامحلول بر عملکرد، خصوصیات لاشه و دستگاه گوارش در جوجههای گوشتی از سن 1 تا 21 روزگی انجام شد. تعداد 660 قطعه جوجه گوشتی نر بهصورت آزمایش فاکتوریل ۳×2 با شش تیمار و پنج تکرار استفاده شدند. عوامل مورد مطالعه شامل دو اندازه ذرات ذرت (2 و 6 میلیمتر) و سه منبع فیبر نامحلول (لیگنوسلولز، پوسته برنج و پوسته آفتابگردان) بودند. اثر اصلی منبع فیبر بر افزایش وزن بدن و ضریب تبدیل غذایی، معنیدار بود بهطوریکه پرندگان تغذیهشده با جیره حاوی پوسته آفتابگردان، افزایش وزن بیشتر (01/0>P) و ضریب تبدیل غذایی بهتری (01/0>P) نسبت به پرندگان تغذیهشده با دو منبع دیگر فیبر داشتند. بهکاربردن لیگنوسلولز (05/0>P)و اندازه ذرات ریز (01/0>P) منجر به افزایش وزن لاشه، بهترتیب در مقایسه با دو منبع دیگر فیبر و اندازه ذرات درشت گردید. وزن روده کوچک در پرندگانی که جیره حاوی لیگنوسلولز دریافت کردند بالاتر از دو منبع دیگر فیبر بود (01/0>P). تیمارهای آزمایشی تأثیر معنیداری بر طول قسمتهای مختلف روده باریک نداشتند. آسیاب درشت ذرت و بهکاربردن پوسته آفتابگردان،pH سنگدان را به ترتیب در مقایسه با ذرات ریز و لیگنوسلولز کاهش داد (05/0>P). وزن سنگدان در پرندگانی که با جیره حاوی لیگنوسلولز و پوسته آفتابگردان تغذیه شدند بالاتر (01/0>P) از جوجههای تغذیهشده با جیره حاوی پوسته برنج بود. براساس نتایج این تحقیق، به کار بردن 3 درصد پوسته آفتابگردان در جیره از طریق بهبود وزن و کاهش pH سنگدان، عملکرد رشد را بهبود میدهد و میتواند در صنعت جوجههای گوشتی بهصورت کاربردی مورد استفاده قرار گیرد.

بررسی کارایی استراتژیهای بهکاررفته برای کنترل بیماری BVD در نژاد هلشتاین
این پژوهش بهمنظور بررسی شیوع اسهال ویروسی گاوان (BVD) و نحوه عملکرد اقدامات صورتگرفته در جهت کنترل این بیماری طی 50 سال اخیر در برخی از استانهای کشور جهت مبارزه با این بیماری انجام شد. بدین منظور، از 500 رأس گاو هلشتاین متعلق به سه گاوداری صنعتی بسیار موفق در سه استان تهران، اصفهان و قزوین بهصورت تصادفی نمونه خون تهیه و با تکنیک الایزا تیتر آنتیبادی و آنتیژن اندازهگیری شد. همچنین بهمنظور اطمینان از حضور و تعیین تیپ ویروسها، تمامی نمونههای مثبت از طریق تکنیک مولکولی RT-PCR مورد سنجش قرار گرفتند. سپس دستورالعملهای کنترل BVD این واحدهای صنعتی بزرگ بررسی شد. نتایج این تحقیق نشان داد میزان الایزا Ab-/Ag-،Ab+/Ag-،Ab-/Ag+ و Ab+/Ag+ بهترتیب 2/10، 8/78، 2/7 و 8/3 درصد بوده است. شیوع آلودگی آنتیبادی و آنتیژن در مجموع سه گاوداری بهترتیب حدود 80 و 11 درصد گزارش شد که از این میان حدود 8/2 درصد حیوانات، عفونی مولد(PI) بودند و تفاوت معنیداری بین سه گاوداری مشاهده نشد. همچنین نتایج RT-PCR نشان داد همه نمونههای مثبت از نوع تیپ 1 ویروس بوده و تیپ 2 در این پژوهش گزارش نشد. نتایج این تحقیق با توجه به شیوع بالای بیماری نشان میدهد، پروتکلهای بهکاررفته جهت کنترل این بیماری از کارایی مناسب برخوردار نبودهاند. بنابراین بهنظر میرسد مهمترین اقدام جهت مبارزه جدی با این بیماری علاوه بر اعمال اصول امنیت زیستی، استفاده از سیاستهای منسجم کلان مدیریتی در سطح ملی خواهد بود.
