Among the major genes, leptin is one of the largely effective ones the polymorphism of which is closely related with important economic traits in livestock. Throughout this study, 100 blood samples were randomly collected from Naeini sheep breed. DNA was extracted using modified salting out method and Polymerase Chain Reaction employed for amplification of a fragment of 275 bp length from exon 3 of leptin gene. The two methods of PCR-SSCP and DNA sequencing were employed for identification of different genotypes. Results showed a single nucleotide mutation of A to C in the nucleotide position of 49 (in the amplified fragment) indicating a type of silent mutation. The frequency of each A and C alleles and of the two AA and AC genotypes were determined at 0.73 and 0.27, respectively. In the course of this research work, the observed effect on growth and wool production traits was also evaluated. The birth weight in sheep with AC genotype was higher than that in animals with BB genotype (P< 0.01), but there was no other significant effect of the observed genotypes on the other studied traits. DNA به روش نمکی بهینه یافته استخراج و یک قطعه 275 جفت بازی از اگزون 3 ژن لپتین با استفاده از واکنش زنجیره پلی مراز تکثیر شد. به منظور تعیین ژنوتیپ های مختلف از دو روش PCR-SSCP و توالی یابی DNA استفاده شد. نتایج این مطالعه یک جهش تک نوکلئوتیدی A به C در موقعیت باز 49 قطعه تکثیری در این پژوهش (مطابق با موقعیت بازی 200 توالی ثبت شده EF534370.1)که از نوع جهش خاموش بود را نشان داد. فراوانی هر یک از آلل های A و C و فراوانی دو ژنوتیپ AA و AC به ترتیب برابر با 0.73 و 0.27 برآورد شد. در این پژوهش، اثر SNP مشاهده شده روی صفات رشد و تولید پشم مورد بررسی قرار گرفت. وزن تولد در گوسفندان با ژنوتیپ AC به طور معنی داری بیشتر از گوسفندان با ژنوتیپ AA بود (p<0.01)، اما اثر ژنوتیپ های مشاهده شده روی دیگر صفات مورد مطالعه معنی دار نبود. می توان نتیجه گیری نمود که با انجام تحقیقات بیشتر در تحقیقات بعدی با اندازه نمونه های بزرگتر، احتمالا بتوان از این جایگاه نشانگری و یا دیگر جایگاه های مرتبط به ژن لپتین در برنامه های انتخاب بر اساس نشانگرهای مولکولی مبتنی بر DNA بهره گرفت.}, keywords_fa = {Naeini sheep,Leptin gene,Polymorphism,growth traits,Wool production traits,PCR- SSCP,DNA sequencing}, url = {https://ijas.ut.ac.ir/article_35562.html}, eprint = {https://ijas.ut.ac.ir/article_35562_a6986a27974d95c3fb53a1b775893d33.pdf} }
